In the present study, we explored the effects of two-day zolpidem (10 μM) treatment on GABA(A) receptors on the membranes of rat cerebellar granule cells (CGCs) using flunitrazepam binding and semi-quantitative PCR analysis.

If functional uncoupling of GABA and benzodiazepine binding sites at GABA(A) receptors is the mechanism responsible for the development of tolerance following long-term administration of classical benzodiazepines, chronic zolpidem treatment may induce tolerance.

Two-day zolpidem treatment of CGCs did not significantly affect the maximum number (B(max)) of flunitrazepam binding sites or the expression of α1 subunit mRNA. However, as shown by decreased GABA flunitrazepam binding, two-day exposure of CGCs to zolpidem caused functional uncoupling of GABA and benzodiazepine binding sites at GABA(A) receptor complexes.

The pharmacological profile of this drug is different from that of classical benzodiazepines, although it acts through benzodiazepine binding sites at GABA(A) receptors. The aim of this study was to further explore the molecular mechanisms of GABA(A) receptor induction by zolpidem.

Effect of zolpidem on gamma-aminobutyric acid (GABA)-induced inhibition predicts the interaction of ethanol with GABA on individual neurons in several rat.

Previous investigations have suggested a relationship between zolpidem binding within specific brain regions and the ability of ethanol or zolpidem to enhance gamma-aminobutyric acid (GABA)-induced inhibition. The purpose of the present study was to extend our electrophysiological analysis to additional brain sites with high levels of zolpidem binding. In the brain regions chosen, red nucleus and globus pallidus, GABA-induced inhibition was shown to be enhanced by either ethanol or zolpidem on some, but not all, neurons.

The patient had a long history of worsening slurred speech, poor balance, frequent falls, and difficulty writing over a six-year period. Her symptoms went undiagnosed for many years, however, as she did not fit the criteria for typical Parkinson disease and was unsuccessfully trialed with carbidopa-levodopa. Her symptoms progressed over this period to the point that she became wheelchair-bound and dependent on a home health aide to complete activities of daily living (ADLs). Eventually, she was diagnosed with PSP based on meeting the official clinical criteria proposed by the National Institute of Neurological Disorders and Stroke (NINDS) and the Society for PSP (SPSP).
